4 ORVOSKÉPZÉS folyóirat szerzõi útmutatója A folyóirat célja: Az 1911-óta megjelenõ Orvosképzés legfontosabb célja a hazai orvoskollégák folyamatos graduális és posztgraduális képzésének támogatása. A lap elsõsorban olyan munkák közlését tartja feladatának, amelyek az orvostudomány egy-egy ágának újabb és leszûrt eredményeit foglalják össze magas színvonalon úgy, hogy azok a gyakorló orvoshoz, szakorvoshoz, klinikushoz és elméleti orvoshoz egyaránt szóljanak. Emellett lehetõség van eredeti közlemények és esetismertetések benyújtására, és az újság a Semmelweis Egyetem szakmai kötelezõ szinten tartó tanfolyamok elõadási összefoglalóinak is teret ad. Az eredeti közlemények a rendszeres lapszámokban, vagy a témához kapcsolódó tematikus lapszámokban kapnak helyet. Fontos feladatunknak tartjuk, hogy rezidens kollégák tollából származó esetismertetéseket is közöljünk, melyeket mentori ajánlással kérünk benyújtani. A beadott dolgozatokat a szerkesztõbizottság elõzetes bírálatra adja ki, és a kézirat közlésére a bírálat eredményének függvényében kerül sor. Tudományos dolgozat benyújtására az alábbiak szerint van lehetõség: Esetismertetés (case report) Fiatal doktorok (PhD) tudományos beszámolója, új eredményeinek összefoglalása (nem tézisek vagy doktori értekezések!) Klasszikus összefoglaló közlemény az elméleti és klinikai orvostudomány bármely területérõl, a legújabb irodalmi eredmények felhasználásával Update jellegû közlemény, azaz nem egy téma kidolgozása, hanem adott szakterület legújabb tudományos eredményeinek összefoglalása Elõadási összefoglaló (a tanfolyamszervezõk felkérése alapján) A kézirat: A tudományos közleményeket elektronikusan, Word dokumentum formátumban kérjük eljuttatni a szerkesztõségbe. Az illusztrációkat, ábrákat és táblázatokat külön file-ként kérjük elküldeni. Az ábrák címeit és az ábramagyarázatokat a Word dokumentumban külön on kell feltüntetni, az ábra/táblázat számának egyértelmû megjelölésével. A digitális képeket minimum 300 dpi felbontásban kérjük, elfogadunk tif, eps, illetve cdr kiterjesztésû file-okat. A kézirat elfogadása esetén az ábrákat a szerkesztõség nyomtatott formában is kéri elküldeni. Az orvosi szavak helyesírásában az Akadémia állásfoglalásának megfelelõen, a latinos írásmód következetes alkalmazását tekintjük elfogadottnak. Magyarosan kérjük írni a tudományágak és szakterületek, a technikai eljárások, mûszerek, a kémiai vegyületek neveit. A szerkesztõk fenntartják maguknak a stiláris javítás jogát. A mértékegységeket SI mértékrendszerben kérjük megadni. A kézirat felépítése a következõ: (1) cím, (2) magyar összefoglalás, kulcsszavakkal, (3) angol összefoglalás (angol címmel), angol kulcsszavakkal, (sorrendben): magyar cím, angol cím, (4) rövidítések jegyzéke (ha van), (5) szöveg, (6) irodalomjegyzék, (7) ábrajegyzék, (8) táblázatok, (9) ábrák. Az számozást a címtól kezdve kell megadni és az egyes felsorolt tételeket külön lapon kell kezdeni. (1) A címon sorrendben a következõk szerepeljenek: a kézirat címe, a szerzõk neve, valamint a szerzõk munkahelye, a kapcsolattartó szerzõ pontos elektronikus és postai címének megjelölésével. (2 3) Az összefoglalást magyar és angol nyelven kell beküldeni, külön akon, a következõ szerkezet szerint: Bevezetés ( Introduction ), Célkitûzés ( Aim ), Módszer ( Methods ), Eredmények ( Results ) és Következtetések ( Conclusions ) lényegre törõ megfogalmazása történjék. A magyar és az angol összefoglalások terjedelme külön-külön ne haladja meg a 200 szót (kulcsszavak nélkül). A témához kapcsolódó, maximum 5 kulcsszót az összefoglalók án, azokat követõen kérjük feltüntetni magyar és angol nyelven. (4) A kéziratban elõforduló, nem általánosan elfogadott rövidítésekrõl külön jegyzéket kell készíteni abc-sorrendben. (5) A szövegtörzs szerkezete világos és az olvasó számára átlátható legyen. Eredeti közlemények esetén a Bevezetõ -ben röviden meg kell jelölni a problémafelvetést, és az irodalmi hivatkozásokat a legújabb eredeti közleményekre és összefoglalókra kell szûkíteni. A Módszer részben világosan és pontosan kell leírni azokat a módszereket, amelyek alapján a közölt eredmények születtek. Korábban közölt módszereket esetén csak a metodika alapelveit kell megjelölni, megfelelõ irodalmi hivatkozással. Klinikai vizsgálatoknál a kézirathoz csatolni kell az illetékes etikai bizottság állásfoglalását. Állatkísérletek esetén a Magyar Tudományos Akadémia Egészségügyi Tudományos Tanács állatkísérletekre vonatkozó etikai kódexe érvényes, melyre a metodikai részben utalni kell. A statisztikai módszereket és azok irodalmát is meg kell adni. Az Eredmények és a Megbeszélés részeket világosan kell megszerkeszteni. Referáló közlemények benyújtása esetén a szövegtörzs altémákra osztható, melyeket alcímek vezessenek be. Összefoglaló referátumoknál a szövegtörzs terjedelme ne haladja meg a karaktert (szóközzel), eredeti közleménynél (klinikai, vagy kísérletes) ne haladja meg a karaktert (szóközzel), esetismertetésnél ne haladja meg a karaktert (szóközzel), elõadási összefoglaló esetén pedig ne haladja meg a 8000 karaktert (szóközzel). Irodalom: a hivatkozásokat (maximum 50, elõadási összefoglalónál maximum 10) a szövegben való megjelenés sorrendjében tüntessék fel. A szövegben a hivatkozást a sorszáma jelöli. Hivatkozás cikkre: sorrendben: szerzõk neve (6 szerzõ felett et al./és mtsai), cikk címe, folyóirat neve (Index Medicus szerint rövidítve), év; kötetszám:elsõ-utolsó. Példa: 1. Philadelphia, Lipincott-Raven, 1996; Ábrajegyzék: a megjelenés sorrendjében, arab számmal sorszámozva egymás alatt tartalmazza az ábra címét és alatta rövid és lényegre törõ ábramagyarázatot Táblázatok: külön-külön lapokon kérjük, címmel ellátva és arab számmal sorszámozva. Törekedjenek arra, hogy a táblázat könnyen áttekinthetõ legyen, ne tartalmazzon zavaróan sok adatot. Ábrák: külön-külön lapokon kérjük. Csak reprodukálható minõségû ábrákat, fényképek küldését kérjük (min. 300 dpi felbontásban), a korábban megjelölt file formátumokban. A kézirat elfogadása esetén a nyomtatott ábrát kérjük beküldeni a szerkesztõségbe és az ábra hátán puha ceruzával kérjük jelölni a szerzõ nevét, arab számmal az ábra sorszámát és a vertikális irányát. A formai hiányossággal beküldött kéziratokat nem tudjuk elfogadni. A gyors lektori és korrektúrafordulók érdekében kérjük a legbiztosabb levelezési, illetve címet, telefon- és faxszámot megadni. Elfogadás esetén külön levélben kérjük jelezni, hogy a szerzõk a közleménnyel egyetértenek (és ezt aláírásukkal igazolják), valamint lemondanak a folyóirat javára a kiadási jogról. Írásbeli engedélyt kérünk mellékelni a már közölt adat/ábra felhasználása, felismerhetõ személy ábrázolása, szerzõnek nem minõsülõ személy nevének említése/feltüntetése esetén. A szerkesztõség az általa felkért szakértõk személyét titkossággal kezeli. A kézirat tulajdonjoga a megjelenésig a szerzõt illeti meg, a megjelenés napján tulajdonjoga a kiadóra száll. A megjelent kéziratok megõrzésére szerkesztõségünk nem tud vállalkozni. 6 ORVOSKÉPZÉS Dear Students, dear Audience, It serves for my great pleasure to welcome you all to the Semmelweis International Students Conference. I am delighted to have the possibility to open a scientific meeting uniting 58 young researchers from sixteen foreign universities of different parts of the world (Catholic University of the Sacred Heart, Rome; Karolinska Institutet, Stockholm; Medical and Pharmaceutical University of Târgu Mureº, Romania; Medical University of Warsaw, Poland; Moscow State University of Medicine and Dentistry, Russia; Novosibirsk State Medical University, Russia; Omsk State Medical Academy, Russia; Oporto University, Portugal; Peoples Friendship University of Russia; Pirogov Russian National Research Medical University; Second Univerisity of Naples - School of Medicine and Surgery, Italy; Siberian State Medical University, Russia; South Ural State Medical University, Russia; Students' Society of McGill University (SSMU), Canada; Tver State Medical Akademy, Parma; Italy; University of Edinburgh, United Kingdom). I believe that Budapest is just about to be the perfect place for such an occasion. This year, for the very first time, under the auspices of the Semmelweis International Students Conference, the Semmelweis Students Scientific Association also organizes three practical competitions: a Surgery Practical Competition, János Kabay Pharmaceutical Practical Team Competition, a Dentistry Practical Competition. The purpose of this initiative as well as the above competitions is to enable the participants of the conference to compare not only their theoretical scientific qualification, but also their practical, problem-solving abilities with those of the other students from different countries. The conference welcomes papers in the following sessions: basic sciences, clinical sciences, pharmaceutical and pharmacological sciences, and dentistry. Participants also have the great opportunity to attend the lecture of Prof. Tatiana S. Sorokina, Head of the History of Medicine Department, Peoples' Friendship University of Russia, on the great Russian surgeon, Nikolay Pirogov ( ). Dear participants, I would like to wish you a nice and fruitful time here in Budapest, please take the opportunity to discuss many important questions of your scientific field with your young colleagues and build new partnerships. 7 ORVOSKÉPZÉS DR. MERKELY BÉLA az ORVOSKÉPZÉS felelõs szerkesztõje Tisztelt Elõadók, Résztvevõk és Bizottsági Tagok! Ismét közeledünk az évnek ahhoz a szakaszához, amikor a hallatók egyetemünk Tudományos Diákköri Konferenciáján mérhetik össze tudásukat és oszthatják meg egymással kutatásuk eredményeit. Az elmúlt évi Tudományos Diákköri Konferencia hangulatára nagy örömmel tekinthetünk vissza, amint az Országos Tudományos Diákköri Konferencián nyújtott kiváló eredményekre is. Ezekre alapozva õszintén bízom az idei konferencia sikerességében évi konferenciánkra 358 elõadással jelentkeztek a hallgatók. Az elõzetes bíráló bizottság szakmai véleményezése alapján a Tudományos Diákköri Tanács az elõadások több mint 99%-át fogadta el. A tudományos ülés létrejöttének jelentõs támogatásáért a Richter Gedeon Nyrt. a Semmelweis Egyetem Tudományos Diákkörének Kiemelt Támogatója címet kapta meg. Idén is tovább erõsödnek nemzetközi kapcsolataink. Ebben az évben a Tudományos Diákköri Tanács kezdeményezésére a konferencián résztvevõ hallgatóknak három gyakorlati versenyen is lehetõségük nyílik a nemzetközi TDK konferencia keretében összemérni tudásukat: a Sebészeti Gyakorlati Versenyen egyénileg, a Kabay János Gyógyszerészeti Gyakorlati Versenyen 3-5 fõs csapatokban, illetve a Fogászati Gyakorlati Versenyen egyénileg. Az újító kezdeményezés és az adott versenyek célja, hogy az elméleti megméretetés mellett a konferencia résztvevõi gyakorlati tudásukat is összeméressék külföldi kollégáikkal. Az egyetemi konferenciánkon kiváló teljesítményt nyújtó elõadóknak lehetõsége nyílik a XXXII. Jubileumi Országos Tudományos Diákköri Konferenciára részvételt nyerni ben Budapest ad majd otthont az Orvos- és Egészségtudományi Szekciónak. A jövõ évi konferencián egyetemünk jelentõsen több hallgatójának lesz alkalma szerepelni (169), mint a korábbi években, köszönhetõen a tudományos diákköri mûhelyek folyamatos, magas színvonalú felkészítõ tevékenységének, hallgatóink korábbi Országos Tudományos Diákköri Konferenciákon való kiemelkedõ eredményeinek, továbbá a Tudományos Diákköri Tanács kitartó munkájának. A delegálás során a tavalyi gyakorlatot követve a bíráló bizottságok írásos ajánlást adnak, a végleges döntést pedig a Tudományos Diákköri Tanács hozza meg. Köszönetet szeretnék mondani a bíráló bizottságok elnökeinek, tagjainak, valamint az elõzetes absztraktbíráló bizottság tagjainak, hogy elfogadták felkérésünket, és munkájukkal emelik konferenciánk szakmai színvonalát! Bízom benne, hogy minden résztvevõ számára meghatározó élményt jelent majd a tudományos program, amely rengeteg tapasztalatcserére ad lehetõséget, és hogy az elmúlt év kitartó munkája megtermi méltó gyümölcsét! 16 BASIC SCIENCES I. A mutation in the magnesium channel MagT1 is responsible for an unusual variant of X-linked ATRX syndrome Arduini Daniela V., Luongo Francesca IV. Catholic University of the Sacred Heart Rome arduinidaniela@libero.it, francescaluong@hotmail.it ATRX syndrome results from mutations in the ATRX gene and is mostly characterized by severe intellectual disability (ID) and distinct facial features. A new variant of the syndrome was identified in a family with five males showing ID, dysmorphic features and a range of skin abnormalities (e.g. generalized pruritus, crinkly skin, cutaneous flushing, marked decrease in sensitivity to temperature and/or pain and grooved nails). Of note, the skin/nail pathology is a unique finding that has not been previously reported in ATRX syndrome patients. Genetic characterization of this variant identified a single base pair deletion in the last exon of ATRX and a de novo 0.8kb intronic duplication in MagT1. MagT1 is a plasma membrane Mg2+ channel expressed ubiquitously, with particular abundance in epithelial cells. Genetic changes in MagT1 have been previously reported in a family with X-linked ID and more recently associated with X-linked immunodeficiency. The aim of our work was to assess the functional implications of the novel MagT1 mutation, and verify its causative role in the present ATRX syndrome variant. B lymphocytes from affected, carrier and control subjects were transformed by EBV and maintained in culture as lymphoblast cell lines (LCL). MagT1 mrna expression and protein levels were assessed in LCL by real time PCR and western blot, respectively. Functional analysis of MagT1 channel activity was performed by measuring basal Mg2+ influx ability in Mag-Fluo-4 loaded LCL by spectrofluorimetry. We show that the novel MagT1 mutation resulted in lower gene expression, both at the mrna and protein level, in LCL from patients vs. carrier or control subjects. Furthermore, basal Mg2+ influx ability was also significantly decreased in patient cells, which suggests an impairment of cellular magnesium homeostasis. In conclusion, our data demonstrate that MagT1 disruption functionally contributes to the atypical presentation of ATRX syndrome in the studied family. We believe it could account for the cutaneous pathology, as normal magnesium homeostasis is important for skin health. In addition, this study highlights that multiple mutations can contribute to the phenotypic variability of syndromic ID, and emphasizes the importance of correlating clinical phenotype with genomic and cell function analyses. Mammalian ageing process is associated with elevated levels of oxidative damage of DNA, proteins, and lipids as a result of unbalanced prooxidant and antioxidant activities. Oxidative stress may activate multiple cell signaling pathways including pathways involved in apoptosis. Cardiomyocytes have limited regenerative capacity, such that cell death can result in a loss of viable contractile elements and a decrease in cardiac functional reserve during normal ageing. The goal of the current study was to investigate the age-related dynamics of apoptotic rate and caspase-3 (apoptosis-related cysteine peptidase) activity in left ventricular cardiomyocytes of male Wistar-Kyoto rats. Materials and methods. A total of 24 male Wistar-Kyoto rats of different ages (8 weeks, 15 weeks, 52 weeks) were used in the study. Left ventricular tissue was stored in 10% neutral buffered formalin for histologic studies or frozen for biochemical analyses. Caspase activity was measured in cell lysates according to the manufacturer`s recommendations («Caspase 3 Assay Kit, Colorimetric», Sigma). The terminal deoxynucleotidyl transferase deoxyuridine triphosphate nick end labeling (TUNEL) assay was performed using the «Apo-BrdU-IHC In Situ DNA Fragmentation Assay Kit» (BioVision). Results: Our results show that compared with 8-week old rats (7,50±0,73%), the number of TUNEL positive nuclei appeared to be markedly increased in hearts of 1-year old rats (11,53±0,90%). The measurement of caspase 3 activity in myocardium exhibited age-related decrease of the enzyme activity, which in 1-year-old rats (0,13±0,02) became lower than in 8-week-old animals (0,22±0,03). Conclusion: ageing is accompanied by increased programmed cell death in left ventricular myocardium and caspase independent mechanisms are predominant in its implementation. Its expression is regulated by alternative splicing generating two isoforms: IR-A, which predominates in embryonic tissue, and IR-B, which is highly expressed in adult, insulin-responsive tissues (skeletal muscle, liver, and adipose tissue). The aberrant INSR expression detected in DM1 and DM2 muscles tissues, characterized by a relative increase of IR-A vs. IR-B, was pathogenically related to the insulin resistance occurring in DM patients. To assess if differences in the aberrant splicing of INSR could underlie the distinct fiber type involvement observed in DM1 and DM2 muscle tissues, we have used laser capture micro dissection (LCM) and RT-PCR, comparing the alternative splicing of INSR in type I and type II muscle fibers isolated from muscle biopsies of DM1, DM2 patients and controls. In the controls, the relative amounts of IR-A and IR-B showed no obvious differences between type I and type II fibers, as in the whole muscle tissue. In DM1 and DM2 patients both fiber types showed a similar, relative increase of IR-A vs. IR-B, as also evident in the whole muscle tissue. Our data suggest that the distinct fiber type involvement in DM1 and DM2 muscle tissues would not be related to qualitative differences in the expression of INSR. So we decided to explore the connective tissue remodeling in endometriosis. The goal: to study decorin expression in the endometrial ovarian cysts considering monophase low-dose combined oral contraceptives (COCs) administration. Materials and methods. We examined 14 women with ovarian endometrial lesions aged from 21 to 47 years (27,7 ± 5,2 years). COCs during 6 months before the surgery used 5 patients. It was performed routine surgical treatment: 13 women laparoscopic cyst husking and one case Pfannenshtil laparotomy, supravaginal hyster- and adnexectomy. For two-step streptavidin-biotin immunohistochemical analysis, 5 mkm paraformaldehyde-fixed, paraffin-embedded samples of endometrial cyst were examined for dermatan sulfate proteoglican decorin expression. A quantitative analysis of immunohistochemical reaction products, visualized by diaminobenzidine (DAB), was performed using a microscope Axio Scope.A1, camera AxioCam MRc5 and the software ZEN blue (C. Zeiss). For the further processing of the data the program Statistica 6.0 was used. Results. The 2nd degree of ovarian endometriosis was established in 6 (42.9%), the 3rd degree - in 8 (57.1%) patients. Endometrial cysts were characterized by almost total endometrial epithelial desquamation and high cellular collation of cytogenic stroma. The COCs administration promoted the fibrosis development with cellular infiltration reduction and synthesis of fibrillar component of connective tissue. Cytogenic stroma of ovarian endometrial cysts was characterized by more prominent decorin expression with the intracellular concentration of the DAB-positive products compared the samples of patients treated COCs before the surgery (p = 0.047). The fibrous tissue of endometrial cyst was characterized by focal and diffuse extracellular decorin expression associated with the collagen fibers localization. Simultaneously the fibrous parameters had virtually no differences depending on COCs administration, with statistically insignificant tendency to increase the numerical indices (p = 0,055). Conclusion. Considering the presence of fundamental analogies between the endometrial disease and cancer, we have particularly demonstrated the proteoglycan decorin expression in surgical endometrial cysts depending the COCs administration. 18 BASIC SCIENCES I. 110 Detailed hemodynamic characterization of athlete s heart using left ventricular pressure-volume analysis in a rat model Kellermayer Dalma IV., Birtalan Ede VI., Semmelweis University dalmakeller@gmail.com; birtalanede@gmail.com Long-term exercise training is associated with characteristic structural and functional changes of the myocardium, resulting in a condition called athlete s heart. Although exercise-induced left ventricular (LV) hypertrophy has been investigated by several groups in animal models, a detailed hemodynamic characterization is not available. We aimed at understanding the functional and morphological changes in the heart following a three-month-long training period in a rat model. Athlete s heart in rats was induced by swim training. The swimmer group was exposed to 200 min/day exercise for 12 weeks. The control group swam only 5 min/day. Following the training period we assessed LV hypertrophy with echocardiography and performed LV pressure-volume (P-V) analysis with a pressure-conductance microcatheter to investigate in vivo cardiac function. Finally, cardiac tissue histology was examined. Echocardiography showed hypertrophy which was confirmed by LV wall-thickness (LV mass index: 2.41±0.08 vs. 2.03±0.08g/kg, p=0.006) and heart weight data (1.73±0.08 vs. 1.41±0.03g, p<0.001). Histology also verified LV hypertrophy (cardiomyocyte diameter: 17.1±0.6 vs. 14.4±1.0µm, p=0.007). We found unaltered heart rate, arterial pressure and LV end-diastolic volume along with decreased LV end-systolic volume, thus increased stroke volume (175±8 vs. 145±8µl, p=0.017) and ejection fraction (73±1 vs. 64±2%, p<0.001) in the swimmers by invasive hemodynamic measurements. The P-V-loop-derived sensitive, loadindependent contractility indexes, such as slope of end-systolic P-V relationship or preload recruitable stroke work (77±7 vs. 54±5Hgmm, p=0.012) were significantly increased. The observed improvement of ventriculoarterial coupling along with increased LV stroke work (15±1 vs. 11±1Hgmm ml, p=0.003) and mechanical efficiency (92±9 vs. 60±6mW, p=0.007) reflect improved mechanoenergetics of athlete s heart. Despite the significant hypertrophy, we observed unaltered LV stiffness and improved LV active relaxation (Tau: 9.6±0.3 vs. 10.9±0.3ms, p=0.006). According to our knowledge this is the first study that characterizes functional changes and hemodynamic relations in exercise-induced cardiac hypertrophy. Today about 30% of couples meet difficulties with conception of the child, to 80% of cases are caused by chronic endometritis (CE) existence. Difficulties of diagnostics of CE are bound to impossibility to make the diagnosis on the basis of general clinical methods of research. Today the gold standard is the morphological method with detection of plasmocytes in a stroma of endometrium, difficulties are bound to a choice of criterion of diagnostics and variability of an assessment of a preparation different researchers. Solution - creation of a marker way of diagnostics which would allow objectively and to draw in a short space of time the conclusion about existence. Research objective: assessment of efficiency of various ways of diagnostics of CE. Materials and methods. For research 95 samples of a tissue an endometrium were taken, from them 50 specimens were stained in the traditional way (a hematoxylin and eosine) and 45 with immunohistochemical detections of CD-138 of positive cells in a stroma an endometrium. 4 pathologists were offered to make the diagnosis with use of traditional approach and with application of a marker method of detection of CD138-positive plasmocytes. At data processing the method of an assessment of a consent of experts (Cohen s kappa) is used. Results. In a course to an assessment rather frequent discrepancy of opinion of the experts, caused by variability of identification of plasmocytes was taped by a traditional method, and at an assessment of the preparations painted by a method of an immunohistochemistry, in most cases the opinion of pathologists coincided. The Kappa coefficient made at a traditional method - 0,42 and with CD138-0,86. Conclusion. Consent coefficient between researchers at an immunohistochemical method 0,86 that corresponds to very good level of a consent, at a traditional method 0,46, the average level of a consent. Disadvantage of an immunohistochemical method is possibility of two-dimensional reactions, when are painted not only plasmocytes, but also epithelial cells. Despite it, an immunohistochemistry a convenient and reliable way of detection of endometrial plasmocytes at CE. Peoples Friendship University of Russia The cell wall of pathogenic yeast of the genus Candida consist of three components: mannan, glucan and chitin. Chitin in the cell wall of yeast is generally present in the amount of up to 10 %. In our previous study, lyticase, the effective enzyme of soil bacteria Cellumonans ceulllans was proven to be successful in causing lysis of these layers and enhanced the effect of various antimycotic medications [Sachivkina et al., 2013]. The next step in our study was the search for analogous producers of microbial origin that could cause the lysis of the chitin layer of the fungal cell wall with a subsequent goal of creating a new complex antifungal medication of biological origin, acting simultaneously on all three layers of the cell wall of Candida. Chitin is one of the most widespread bio-polymers. Its modification and degradation occurs in nature through various chitinolytic enzymes. One of the key aspects among the various areas of practical applications of these enzymes is the development of biologically-based medical control of fungal pathogens adversely affecting humans. These types of fungicides are environmentally safe biological agents and are cheap to produce. In this study we explored the antagonistic interaction of chitinases with yeast-like fungi of the genus Candida. We used three different strains of microorganisms: Bacillus licheniformis; Cellulomonas cellulans; Aspergillus foetidus. Our technical objective was to reduce the duration of incubation and to maximize the output of chitinase, as well as to simplify of the media. For successful cultivation of the microorganisms, the following media was applied (g/l of dry substrate): yeast S. cerevisiae ; 1.0 L water, ph 8.0. The cells of S. cerevisiae used in the method described contain molecules of chitin, which serves as a direct inducer of chitinase production. Optimal culture growth conditions were identified for all three strains producing the enzyme in maximum quantity. The germination of the filamentous forms of Candida albicans treated with culture fluid from Aspergillus foetidus was inhibited by 35-50% on the second day of incubation. Our next step is the isolation and characterization of purified chitinase, determining its activity in unit/ ml of medium, as well as the degree of suppression of fungal growth in vitro and in vivo. Catholic University of the Sacred Heart Rome l.iaia@hotmail.it; domenico.damario@gmail.com Intracoronary infusion of Cardiac Stem Cells (CSCs) isolated from right atrial appendage, resected during CABG is effective in improving left ventricular systolic function and reducing infarct size in patients with ischemic heart failure. We sought to develop in vivo, a minimally invasive and reproducible methodology for the isolation and expansion of CSCs from endomyocardial biopsies (EMB) in humans. In our center, 35 patients with clinically suspected myocarditis (n=20) and/or cardiomyopathy of nonischemic origin including those with infiltrative or connective tissue disease (n=15) underwent either selective LV-EMB (n=26), selective RV-EMB (n=9), or biventricular EMB (n=5) after coronary angiography. Cardiovascular magnetic resonance was performed in all patients. Moreover, EMB were drawn according to intracardiac ecocardiographic imaging and under 3D-electroanatomic guidance (3-EAM). Samples not used for diagnostic purposes were enzymatically dissociated, and the unfractionated cell population was expanded for 22±4 days. In all patients considered, CSC were successfully isolated. After expansion cells were sorted for c-kit: c-kit positive CSCs were obtained in all patients considered and characterized. At P6, 70% (69±5.3%) of CSCs still expressed c-kit in culture and were negative for markers of hematopoietic and mesenchymal lineage. Only a small fraction of CSCs were positive for markers of myocyte, endothelial or smooth muscle cell lineage. Population doubling time was calculated and averaged 27.5±2.2 hours. Additionally at P6, CSC were exposed to a pulse of bromodeoxyuridine and analyzed 12 hours later: 8.1±1.3% CSC were positive for BrdU. Telomere length was measured in CSCs by flow-fish: telomeres varied from 6.3 to 7.8 kbp, far from telomere lengths associated with replicative senescence and growth arrest. Consistently, only 3.1±1.6% of CSCs were positive for p16ink4a. In the present study, we established the conditions for the isolation and expansion of c-kit-positive CSCs from EMB. 3-EAM guidance may allow to accurately identify low voltage areas corresponding to areas of active inflammation or scar tissue, thus improving safety and harvesting performance even in the setting of non-ischemic cardiomyopathies. Introduction: Notch receptors constitute a simple yet versatile signalling pathway. Notch3 is primarily expressed in vascular smooth muscle cells and plays an important role in the development of the vascular system. The signalling pathway is also important in a number of small vessel diseases, including pulmonary arterial hypertension and CADASIL; a hereditary stroke disorder. Connections have been made between the pathogenic mechanisms underlying these diseases and the altered vascular myogenic tone and response observed in Notch3 deficient mice. Aims: To determine whether Ca 2+ -signalling, an important agent in mediating a myogenic response, is altered in Notch3-/- cells compared to Notch3+/- and wild-type cells. Materials and methods: Vascular smooth muscle cells isolated from aortas of Notch3-/-, Notch3+/- and wild-type mice were cultured and studied using confocal microscopy with Fluo-4 as a fluorescent marker of free cytoplasmic intracellular Ca 2+. Thapsigargin, a SERCA-channel blocker, and extracellular Ca 2+ were added to the cells to study changes in intracellular Ca 2+ -concentrations. Results: The experiment did not show statistically significant differences in Ca 2+ -release from the sarcoplasmic reticulum after the addition of Thapsigargin in Notch3-/- cells compared to Notch3+/- and wild-type cells. While the degree of variability between studies was high, a trend of lower Ca 2+ release from Notch3-/- cells was observed. Conclusions: The findings indicate that the Ca 2+ -storage capacity could be reduced in Notch3-/- cells, but further studies are needed as the results from this study are inconclusive. Furthermore, when the electron transport chain is dysfunctional, provision of succinyl-coa by the alphaketoglutarate dehydrogenase complex (KGDHC) is crucial for maintaining the function of succinyl-coa ligase yielding ATP, preventing the adenine nucleotide translocase from reversing. We addressed the source of the NAD+ supply for KGDHC under anoxic conditions and inhibition of complex I. Using pharmacologic tools and specific substrates and by examining tissues from pigeon liver exhibiting no diaphorase activity, we showed that mitochondrial diaphorases in the mouse liver contribute up to 81% to the NAD+ pool during respiratory inhibition. Under these conditions, KGDHC s function, essential for the provision of succinyl-coa to succinyl-coa ligase, is supported by NAD+ derived from diaphorases. Through this process, diaphorases contribute to the maintenance of substrate-level phosphorylation during respiratory inhibition, which is manifested in the forward operation of adenine nucleotide translocase.
